About TB-500
What it is
TB-500 is a synthetic version of a short region of Thymosin Beta-4, a protein found in almost every human cell and in high concentration in wound fluid and platelets. Rather than the whole 43-amino-acid protein, this is the active binding fragment, which makes it cheaper to synthesise and easier to keep in solution.
How it is understood to work
Its defining property is binding to G-actin. Actin is the protein that gives a cell its skeleton and lets it change shape and move, and by sequestering the free monomer form, this fragment influences how readily actin filaments assemble and disassemble. Cell motility depends on exactly that cycle, which is why it appears so often in migration and remodelling research.
What research has looked at
Laboratory work has examined cell migration, endothelial cell behaviour, corneal and dermal wound models, and cardiac tissue models. Interest largely follows from the actin-binding mechanism rather than from any single dramatic finding. Handling and storage
Lyophilised and hygroscopic — it will pull moisture from the air, so let the vial reach room temperature before opening or you will condense water into the powder. Reconstitute gently down the side of the vial rather than directly onto the pellet; the actin-binding region is a flexible chain and vigorous shaking encourages aggregation. Refrigerate once in solution.
