About SS-31 (Elamipretide)
What it is
SS-31, also called elamipretide, is a four-amino-acid aromatic-cationic peptide designed to reach the inner mitochondrial membrane. Getting a molecule to a specific organelle is difficult, and the way this one does it is the notable part.
How it is understood to work
It binds cardiolipin, a phospholipid found almost exclusively in the inner mitochondrial membrane and essential to the structure of the cristae and the function of the electron transport chain. By associating with cardiolipin the peptide concentrates where it is wanted without needing an active transport mechanism, and research has examined whether this stabilises cristae architecture and improves electron transport efficiency.
What research has looked at
Mitochondrial function, cardiolipin biology, and models of mitochondrial myopathy. It has been through clinical trials, with mixed results, and holds no marketing authorisation.
Handling and storage
Lyophilised, reconstitute with bacteriostatic water, refrigerate. Cationic peptides can adsorb to surfaces, so low-binding labware is worth considering for low-concentration work.
